目的 研究不同产地生附子在破格救心汤中双酯型乌头碱质量分数的差异.方法 采用高效液相色谱法,以Technologies Athena C18-WP (4.6 mm×250 mm,5 μm)为色谱柱,甲醇(0.1%三乙胺)-水(体积比70 :30)为流动相,流速为1.0 mL/min,柱温为35℃,检测波长为235 nm.对13个不同产地生附子组成的破格救心汤中双酯型乌头碱的质量分数进行检测,比较不同产地生附子在破格救心汤中毒性成分质量分数的差异.结果 13个不同产地生附子组成的破格救心汤水煎液中3种毒性双酯型乌头碱的质量分数具有明显差异,以四川江油产地的附子总双酯型乌头碱质量分数为标准,与其相近的为陕西汉中、江苏常州的样品;质量分数较高的为云南腾冲、辽宁大连、四川凉山的样品,其中云南腾冲样品约为四川江油的3倍;质量分数较低的为河北石家庄、河南南阳、广西百色的样品.结论 本文可为临床应用破格救心汤时,附子产地选择、剂量使用、安全性评价及临床安全用药提供科学依据.

Objective To investigate the contents of the diester aconitum alkaloids in Poge Jiuxin Decoction with Radix Aconite Lateralis from different regions. Methods High performance liquid chromatography ( HPLC) was used to determine the mass fraction of the diester aconitum alkaloids in Poge Jiuxin Decoction. Its conditions were as follows: Technologies Athena C18-WP ( 4.6 mm×250 mm, 5 μm) column with methanol ( 0.1% triethylamine) and H2O ( 70 :30) as a mobile phase, the flow rate was 1.0 mL/min, the column temperature was 35 ℃ and the detection wavelength was 235 nm. The relationship between toxicity and regions was explored with diester aconitum alkaloids as an indicator. Results The mass fraction of three kinds of diester aconitum alkaloids in the decoction were obviously different between the thirteen different regions of Radix Aconite Lateralis. The mass fraction of total diester aconitum alkaloids in Radix Aconite Lateralis from Jiangyou, Sichuan province, was taken as a standard. The samples from Hanzhong, Shanxi province, and Changzhou, Jiangsu province were similar to the standard, those from Tengchong, Yunnan Province, Dalian, Liaoning province, and Liangshan, Sichuan Province were much higher, and the one from Tengchong, Yunnan province was approximately 3 times the standard, while these from Shijiazhuang, Hebei province, Nanyang, Henan province and Baise, Guangxi Province were much lower than the standard. Conclusion The results could provide scientific reference on relationship among the regions, dosage, clinic drug use and safety evaluation for further application of Poge Jiuxin Decoction.
